Abstract目的: 建立测定马比木根中喜树碱和9-甲氧基喜树碱的高效液相色谱法。方法: 采用5种方法对马比木根中的喜树碱和9-甲氧基喜树碱进行提取; 运用高效液相色谱法,采用Waters Symmetry C_(18)色谱柱(4. 6 mm 250 mm,5 mum),以甲醇-水(50∶ 50)为流动相,流速0. 8 mL·min ~(- 1),检测波长254 nm,柱温26 ℃。结果: 喜树碱和9-甲氧基喜树碱进样量分别在0. 24 ~ 4. 00 mug(r = 0. 9997)和0. 24 ~ 4. 00 mug(r = 0. 9995)范围内线性关系良好; 乙醇溶液室温浸泡提取得到的喜树碱含量(0. 944%)和9-甲氧基喜树碱含量(0. 237%)明显高于其他方法。马比木是目前发现的喜树碱类含量最高的植物。结论: 本文建立的HPLC 检测法简便、准确、灵敏度高,乙醇溶液室温浸泡提取法安全,成本低,提取率高,适用于喜树碱类提取和马比木的质量检测。
