Abstract: The present study compared the efficiency of three cryopreservation strategies for Feizhouyou and Mansailong pomelo cultivars collected from Xishuangbanna, China. The approaches tested were whole-seed desiccation by either silica gel or equilibrium desiccation, and isolated embryonic axis desiccation. Regardless of the desiccation method used, Feizhouyou seeds displayed higher desiccation tolerance and post-thaw viability than Mansailong. In both cultivars, seeds desiccated by silica gel exhibited higher desiccation tolerance and post-thaw viability than those from equilibrium desiccation. Maximum post-thaw emergence in Feizhouyou and Mansailong was 77.5 and 63.3%, respectively. In both cultivars, higher desiccation tolerances and cryotolerances were obtained using embryonic axis cryopreservation than with the whole-seed approaches. Feizhouyou seeds and axes displayed higher emergence percentages before and after freezing than those of Mansailong, confirming the variation observed in desiccation tolerance and cryotolerance between these cultivars. Our results suggest that equilibrium desiccation by saturated salt solutions may not be suitable for cryopreservation of pomelo seeds.
